Abstract   Normal ar B16 tumor-bearing C57BL/6 mice were injected intravenously with 30 ug of polyadenylic. polyuridylic acid [poly(A). poly(U)]. Spleen cells, prepared from these mice 48h later, were cultured in media containing 300 units/ml of recombinant human interleukin-2 (rHIL-2) for 5 days. Cytotoxic activity of cultured cells were measured by a 4h-51Cr release assay using NK-sensitive YAC-1 and NK-resistant L1210 target cells, and their surface phenotypes were determined by an immunofluorescence technique using monoclonal antibodies. The results were compared to those obtained with spleen cells of the same batch before culture. Fresh spleen cells from either normal or tumar-bearing mice showed a cytotoxicity relatively low agajnst YAC-1 and almost null against L1210 cel1s. Nevertheless, spleen cells from poly(A). poly(U)- treated mice of both groups showed a higher activity against YAC-1 cells than those from saline- treated control mice. In contrast, the cultured spleen cells showed significantly increased cytotaxic activity not only against YAC-1 but also against L1210 cells, and these activities were more pronounced in poly(A). poly(U)-treated tumor-bearing mice. Proliferations of spleen cells in culture, measured by the technique of 'H thymidine incorporation, were also significantly increaeed in spleen cells from poly(A). poly(U)-treated mice. Major phenotypes of cultured cells were Thyl.2' asialo GM1'. From these results, it can be concluded that treatment of mice with poly(A). poly(U) has enhanced significantly the in vitro generation of LAK ceils as well as their cytotoxic function against tumor target cells, suggesting that bath LAK and NK cells could be involved as effectors in the mechanisms of anti-tumor activity of poly(A). poly(U).
